Determining Whether Sleep Deprivation Contributes to Locomotor Deficits in Aβ-42 Transgenic Drosophila melanogaster

SOMERS Science Fair  ·  Medicine & Health  ·  Spring 2026 – Fall 2027

Approximately 30% of adults are affected by sleep deprivation, and research suggests that ~15% of Alzheimer's disease cases are linked to sleep disruption. My project investigates whether chronic sleep loss accelerates the onset of Alzheimer-like symptoms — specifically locomotor deficits — in Aβ-42 transgenic Drosophila melanogaster fruit flies.

Using a Power Tower to continuously disrupt sleep and a Negative Geotaxis Assay measured at Days 1, 5, 10, 15, and 20, I compared motor decline between sleep-deprived and non-sleep-deprived transgenic fly groups. The hypothesis: sleep-deprived flies will show locomotor deficits earlier — evidence that insomnia accelerates amyloid-beta accumulation and neurodegeneration.

Key Finding

Sleep deprivation is predicted to increase Aβ-42 plaque build-up in the brain, causing earlier breakdown of motor function — supporting sleep as a critical, modifiable risk factor for Alzheimer's disease.
